چکیده

We present a microfluidic neuron culture platform capable of isolating/guiding CNS axons from neuronal somata for localized manipulation and quantitative analysis of axonal growth. Localized treatment of five extracellular matrices to somata or axons for investigating their effects on axon growth was demonstrated. Quantitative analysis of axon growths in response to different treatment could be performed in significantly reduced time due to the axon guiding feature of the platform. Isolation capability, along with the multi-compartment configuration enabling six different localized axon treatments to be carried out on a single device simultaneously, make this neuron culture platform ideal for screening drugs and molecular factors that promote CNS axon growth.

A Microchip for High-Throughput Axon Growth Drug Screening

It has been recently known that not only the presence of inhibitory molecules associated with myelin but also the reduced growth capability of the axons limit mature central nervous system (CNS) axonal regeneration after injury. Conventional axon growth studies are typically conducted using multi-well cell culture plates that are very challenging to investigate localized effects of drugs and li...

Microfluidic Co-culture Platform for Cns Axon Myelination

A microfluidic compartmentalized co-culture platform for central nervous system (CNS) axon myelination research has been developed. The platform is composed of two compartments connected via arrays of shallow microfluidic channels that function as fluidic and physical isolation barrier between the two compartments. Neurons from E 16-18 rats and oligodendrocytes (OLs) from P1-2 rats were cocultu...

Three Dimensional Co-culture of Neuron and Astrocyte in a Micro-fluidic Deivce

In this paper, we describe a microfluidic platform that enables three dimensional cell culture within defined microenvironments towards a study of axon-glia interaction. The microdevice can offer both 3D axon isolation from cell body and 3D spatial cell separation between neuron and astrocyte. It consists of large fluidic channels for media supply and small channels for collagen filling as a th...

Curcumin protects axons from degeneration in the setting of local neuroinflammation.

Axon degeneration is a hallmark of several central nervous system (CNS) disorders, including multiple sclerosis (MS), Alzheimer's disease (AD) and Parkinson's disease (PD). Previous neuroprotective approaches have mainly focused on reversal or prevention of neuronal cell body degeneration or death. However, experimental evidence suggests that mechanisms of axon degeneration may differ from cell...

Streamlining gene expression analysis: integration of co-culture and mRNA purification.

Co-culture of multiple cell types within a single device enables the study of paracrine signaling events. However, extracting gene expression endpoints from co-culture experiments is laborious, due in part to pre-PCR processing of the sample (i.e., post-culture cell sorting and nucleic acid purification). Also, a significant loss of nucleic acid may occur during these steps, especially with mic...
